【Abstract】AIM:ToexpressrecombinantmouseWnt3asignalproteinwithbiologicalactivity.METHODS:Therecombinanteukaryoticexpressionplasmid,pSecTag2/HygroBWnt3a,wastransfectedintoNIH3T3cellsbyliposome,thentheexpressedproteinwasdetectedbyWesternBlot.ThesaturationdensityandthecapabilityofsuppressingapoptosisinWnt3a/NIH3T3cellswereevaluated.RESULTS:TheWnt3asignalproteinwasstablyexpressedinWnt3a/NIH3T3cells.ThesaturationdensityandthecapabilityofsuppressingapoptosisinWnt3a/NIH3T3cellsweremarkedlyincreased.CONCLOUSION:WesuccessfullyconstructtherecombinantWnt3asignalproteinwithbiologicalactivity.

【摘要】目的:表達具備生物活性的重組小鼠Wnt3a信號蛋白.方法:應用脂質體轉染試劑將重組真核表達載體pSecTag2/HygroBWnt3a轉染并篩選穩定表達的NIH3T3細胞,WesternBlot鑒定重組Wnt3a蛋白的表達,并對Wnt3a/NIH3T3細胞的融合密度及抗凋亡能力給予檢測.結果:Wnt3a信號蛋白在Wnt3a/NIH3T3細胞中獲得穩定表達,Wnt3a信號蛋白能夠明顯提高NIH3T3細胞的融合密度及抗凋亡能力.結論:在NIH3T3細胞中表達的重組Wnt3a信號蛋白具備生物活性.

【Abstract】AIM:Toobservetheeffectsofoctylphenol(OP)oncellcycleandcyclinexpressionofMDAMB231breastcancercellsandtoprobethemolecularmechanismsofOPinhibitingMDAMB231breastcancercellproliferation.METHODS:Thechangesofcellproliferation,cellcycle,cyclinD1mRNA,cyclinD3mRNA,CDK2mRNA,CDK4mRNAandcyclinD1proteinexpressionsofMDAMB231cellstreatedwithOPwereobservedbyusingMTTtest,flowcytometry,immunocytochemistryandRTPCR.RESULTS:WhenMDAMB231breastcancercellsweretreatedwith4or8μmol/LOPfor48h,thecellinhibitionrateswere(17.13±4.1)%,(40.25±8.7)%,andthefluorescencevaluesofcyclinD1were55.1±10.2,41.4±11.2,significantlydecreasedascomparedwithcontrolgroup(89.9±11.2,P<0.05).WhenMDAMB231breastcancercellsweretreatedwith8μmol/LOPfor24and72h,thecellratiosintheG0/G1phasewere(67.0±17.6)%and(44.4±11.9)%,significantlyincreasedascomparedwithcontrolgroup[(46.6±12.8)%at24h,(15.3±3.1)%at72h,P<0.05].OPinhibitedthemRNAexpressionsofcyclinD1,cyclinD3,CDK2andCDK4andtheproteinexpressionofcyclinD1.CONCLUSION:OPcaninhibittheproliferationofMDAMB231breastcancercells,whichmayberelatedtodownregulatingtheexpressionsofcyclinD1,cyclinD3,CDK2andCDK4mRNAandcyclinD1protein.

【摘要】目的:觀察辛基酚(OP)對細胞周期及周期蛋白表達的影響,探討OP抑制MDAMB231乳腺癌細胞增殖的可能分子機制.方法:以MTT試驗、流式細胞分析、免疫細胞化學和RTPCR等方法觀察OP對MDAMB231乳腺癌細胞增殖、細胞周期、CDK2,CDK4,cyclinD1和cyclinD3表達的影響.結果:4,8μmol/LOP作用MDAMB231乳腺癌細胞48h時,其抑制率為(17.1±4.1)%,(40.3±8.7)%,cyclinD1表達量熒光值為55.1±10.2,41.4±11.2,比對照組(89.9±11.2)明顯降低(P<0.05).8μmol/LOP作用MDAMB231乳腺癌細胞24h和72h,G0/G1期細胞為(67.0±17.6)%和(44.4±11.9)%,G0/G1期細胞比對照組[24h,(46.6±12.8)%;72h,(15.3±3.1)%]明顯增高(P<0.05).OP導致MDAMB231乳腺癌細胞中CDK2,CDK4,cyclinD1和cyclinD3mRNA的表達降低,且cyclinD1蛋白的表達也降低.結論:OP能抑制MDAMB231乳腺癌細胞增殖,其機制可能與OP能降低乳腺癌細胞中CDK2,CDK4,cyclinD1和cyclinD3mRNA及其蛋白的表達有關.

【摘要】目的:檢測大腸癌中VHLmRNA,FIH1mRNA以及HIF1α蛋白的表達,探討VHL,FIH1對HIF1α蛋白水平的影響.方法:采用RTPCR技術檢測42例大腸癌組織和相應的癌旁正常粘膜組織中VHLmRNA,FIH1mRNA的表達,采用免疫組化SP法檢測HIF1α蛋白在大腸癌和癌旁正常組織中的表達,Spearman相關關系檢驗分析其之間的相關性.結果:大腸癌組織VHLmRNA和FIH1mRNA的表達水平顯著低于癌旁正常組織(t=-10.14,P=0.00;t=-15.26,P=0.00);二者的表達水平隨Dukes分期而降低,DukesA+B期顯著高于DukesC+D期(t=2.84,P=0.01;t=-5.13,P=0.00);它們的表達水平與與腫瘤部位、性別無關.大腸癌組織HIF1α蛋白表達陽性率為69.1%(29/42),在癌旁正常組織中均為陰性;DukesC+D期(80.0%,24/30)顯著高于DukesA+B期(41.7%,5/12),(t=5.89,P=0.02);Spearman相關分析顯示,HIF1α表達水平與VHLmRNA,FIH1mRNA的表達水平顯著負相關(rs=-0.97,P=0.01,rs=-0.66,P=0.01).結論:HIF1α的過表達在大腸癌的發生發展、浸潤轉移過程中起著重要作用,大腸癌組織中VHL以及FIH1的水平下降與HIF1α蛋白水平的升高之間存在密切相關.

【Abstract】AIM:TodetecttheroleandthesignificanceofMMP9inmicroangiogenesisinskinsquamouscellcarcinoma(SCC)andtoobservetherelationbetweentheexpressionofMMP9andmicrovesseldensity(MVD)inSCC.METHODS:ImmunohistochemicalSPstainingwasusedtoexamineMMP9expressionandthechangeofMVDunderlightmicroscopein45casesofSCC,19casesofBowendisease,and10casesofnormalskin.RESULTS:TheexpressionofMMP9inSCCgroupwassignificantlyhigherthanthatinBowendiseasegroup(P<0.05),whilethelatterwasalsosignificantlyhigherthanthatinnormalskingroup(P=0.028,P<0.05).ThelevelofMVDinSCCgroupwassignificantlyhigherthanthatinBowendiseasegroup(P<0.05),andthelatterwassignificantlyhigherthanthatinnormalskingroup(P<0.05).CONCLUSION:MMP9ishighlyexpressedinSCC,andmaybeoneofimportantfactorsinangiogenesisofSCC.

【摘要】目的:探討皮膚鱗狀細胞癌(SCC)發生發展過程中基質金屬蛋白酶9(MMP9)對微血管生成的意義.方法:SCC45例,Bowen病19例、正常皮膚10例進行CD34,MMP9免疫組化染色.檢測MMP9的表達和微血管密度(MVD)的變化,分析MMP9的表達與MVD之間,及它們與SCC臨床病理特征之間的關系.結果:在SCC組中MMP9表達高于Bowen病組(P<0.05),Bowen病組中MMP9表達高于正常皮膚組(P=0.028,P<0.05).在SCC組中MVD值高于Bowen病組(P<0.05),Bowen病組中MVD值高于正常皮膚組(P<0.05).結論:MMP9可能是SCC促進微血管生成因子之一,與SCC的侵襲性相關.
